Role of Glutathione aganist Cytotoxicity of t-Butyl Hydroperoxide in Cultured Liver Cells

Abstract
Free radicals contains an unpaired electron, which are very unstable compounds, are continually generated and eliminated in the biologic systems. The free radicals are so active that they can cause cell damage by lipid peroxidation, protein oxidation and nucleic acid damage. In this study t-butyl hydroperoxide (t-BHP) which can produce intracellular free radicals product of lipid peroxidation, were used for elucidating Glutathione (GSH) role in the mechanisms of cell damage by free radicals as well as in the protection of liver cell by cobalt among metals. The cytotoxicities of t-BHP were showed in dose-dependent matters. The pretreatment with cysteine precursor, oxothiazolidine carboxylate (OTZ) reduced cell damage by t-BHP. Pretreatment with GSH synthesis inhibitor and GSH deplete in the cultured liver cell exposed to t-BHP elevated the cytotoxicity, which suggest that GSH plays an important role in cellular defense against t-BHP. Cobalt reduced the cytotoxicity by t-BHP. These reductions of cytotoxicity were blunted by pretreatment with GSH synthesis inhibitor and GSH deplete, which suggests that GSH play an important role in protection of cells from cytotoxicity of t-BHP. In conclusion, we assume that GSH depletion might play a role in cell damage by free radicals.
